Peptide-based compounds including heteroatom-containing, three-membered rings efficiently and selectively inhibit specific activities of N-terminal nucleophile (Ntn) hydrolases. The activities of those Ntn having multiple activities can be differentially inhibited by the compounds described. For example, the chymotrypsin-like activity of the 20S proteasome may be selectively inhibited with the inventive compounds. The peptide-based compounds include an epoxide or aziridine, and functionalization at the N- terminus. Among other therapeutic utilities, the peptide-based compounds are expected to display anti-inflammatory properties and inhibition of cell proliferation.

La présente invention a trait à des composés à base de peptides comportant des noyaux à trois chaînons contenant des hétéroatomes qui sont inhibiteurs d'activités spécifiques d'hydrolases nucléophiles de N-terminal (Ntn). Les activités de ces Ntn ayant une pluralité d'activités peuvent être inhibées différemment par les composés de l'invention. Par exemple, l'activité de type chymotrypsine du protéasome 20S peut être inhibée de manière sélective avec les composés de l'invention. Les composés à base de peptides comprennent un époxyde ou une aziridine, et la fonctionnalisation au niveau du N-terminus. Parmi d'autres utilisations thérapeutiques, les composés à base de peptides sont prévus pour présenter des propriétés anti-inflammatoires et l'inhibition de la prolifération cellulaire.
