An optical computed-tomography microscope for three-dimensional (3-D) imaging employs tomographic reconstruction for image acquisition. The microscope has an optical scanner to vary an angle at which a light beam passes through a specimen. A method for limited-angle computed-tomography reconstruction applies a transform to produce an image from a number of projections. The image is iteratively feedback-corrected.

La présente invention concerne un microscope de tomodensitométrie optique conçu pour une imagerie en trois dimensions (3D). Ce microscope utilise une reconstruction tomographique pour l'acquisition d'images et comprend un scanneur optique conçu pour faire varier un angle selon lequel un faisceau lumineux passe à travers un échantillon. Cette invention concerne également un procédé de reconstruction par tomodensitométrie à angle limité qui applique une transformation pour produire une image à partir d'un certain nombre de projections.
