Suppression of hematopoietic-specific SH2-containing inositol polyphosphatase (SHIP) activity by genetic and pharmaceutical means is taught for suppression of rejection of, and prevention of graft-versus-host disease in, solid organ allografts or xenotransplants, and histo-incompatible marrow grafts. Also disclosed are methods for the screening of substances and genetic constructs that inhibit SHIP function in mammalian cells, and cell lines and transgenic animals that have the SHIP-/- phenotype.

L'invention concerne la suppression de l'activité de l'inositol polyphosphatase contenant du SH2 (SHIP) à spécificité hématopoïétique par des moyens génétiques et pharmaceutiques afin de supprimer et de prévenir la réaction du greffon contre l'hôte dans les allogreffes ou les xénotransplants d'organes solides et les greffes de moelle histo-incompatibles. L'invention concerne également des méthodes de criblage de substances et de constructions génétiques qui inhibent la fonction SHIP dans les cellules des mammifères ainsi que des lignées cellulaires et des animaux transgéniques qui présentent le phénotype SHIP?-/-¿.
