An extended release dosage form of pharmaceutically active substances that have a water contact angle (.theta.) such that cos .theta. is between +0.9848 and -0.9848 presented as a matrix tablet containing the said pharmaceutically active substances, with/without suitable pharmaceutical excipients in intimate mixture with two groups of intelligent polymers having opposing wettability characteristics, one demonstarting a stronger tendency towards hydrophobicity and the other a stronger tendency towards hydrophilicity, the polymer combination being between the ratios of 1:50 and 50:1 amounts effective to control the release of said pharmaceutically active substances in a mathematically predictable manner, wherein the polymer demonstrating a stronger tendency towards hydrophobicity is not less than 5% wt/wt and preferably between 5-70% wt/wt of the final formulation composition. The intelligent polymers being ethylcellulose (EC) as a more strongly hydrophobic and hydroxyethylcellulose (HEC) and hydroxypropyl methylcellulose (HPMC) as more strongly hydrophilic (the ratio of HEC to HPMC being between 1:100 and 100:1). The matrix tablet is optionally coated with an enteric coat, 0.5% - 15% wt/wt to prevent the initial burst effect seen in such systems and to impart gastrointestinal tract (GIT) "stealth" characteristics especially in the presence of food.

Forme posologique, à libération prolongée, de substances pharmaceutiques - qui donnent un angle de contact avec l'eau (thêta), correspondant à un cos.thêta compris dans un intervalle de +0.9848 à -0.9848 - se présentant en comprimé qui renferme lesdites substances pharmaceutiquement actives, avec ou sans excipients pharmaceutiques appropriés en mélange intime avec deux groupes de polymères intelligents possédant des caractéristiques de mouillabilité opposées, l'un manifestant une forte tendance à l'hydrophobie, et l'autre une forte tendance à l'hydrophilie; la combinaison des polymères se situe entre des rapports de 1:50 et 50:1, quantités permettant de réguler efficacement la libération desdites substances pharmaceutiquement actives, selon un mode prévisible mathématiquement; le polymère manifestant une plus forte tendance à l'hydrophobie représente au moins 5 % en poids/poids, et de préférence entre 5 et 70 % en poids/poids de la composition finale de la formulation. Les polymères intelligents sont l'éthylcellulose (EC), en tant que composante fortement hydrophobe, ainsi que l'hydroxyéthylcellulose (HEC) et l'hydroxypropylméthylcellulose (HPMC), en tant que composantes fortement hydrophiles (le rapport de HEC sur HPMC se situe entre 1:100 et 100:1). Le comprimé est facultativement enrobé d'une couche gastro-résistante, à raison de 0,5 % - 15 % en poids/poids pour empêcher l'effet d'éclatement initial observé avec ce type de système, et pour lui donner un caractère « furtif » au niveau gastro-intestinal, particulièrement en présence d'aliments.
