Novel controlled release pharmaceutical compositions for oral use containing midodrine and/or its active metabolite desglymidodrine. The novel compositions are designed to release midodrine and/or desglymidodrine after oral intake in a manner which enables absorption to take place in the gastrointestinal tract so that a relatively fast peak plasma concentration of the active metabolite desglymidodrine is obtained followed by a prolonged and relatively constant plasma concentration of desglymidodrine. Also disclosed is a method for treating orthostatic hypotention and/or urinary incontinence, the method comprising administration to a patient in need thereof of an effective amount of midodrine and/or desglymidodrine in a composition according to the invention.
L'invention concerne de nouvelles compositions pharmaceutiques à libération contrôlée, à base de midodrine et/ou son métabolite actif, le desglymidodrine. Ces nouvelles compositions sont prévues pour libérer la midodrine et/ou le desglymidodrine après l'administration orale, de telle sorte que l'absorption s'effectue dans le système gastro-intestinal et que l'on obtient un pic de concentration plasmatique relativement rapidement du métabolyte actif, le desglymidodrine. Ce pic est ensuite suivi d'une concentration plasmatique du desglymidodrine prolongée et relativement constante. L'invention a pour objet également un procédé permettant de traiter l'hypotension orthostatique et/ou l'incontinence urinaire. Ce procédé consiste à administrer à un patient une quantité efficace de midodrine et/ou desglymidodrine dans une composition selon l'invention.
