A method is described for obtaining Hoogsteen-paired pyrimidine* purine duplezes, either by heating a triplex to dissociate a Watson-Crick paired pyrimidine strand or by linking two parallel strands at their 5' ends is disclosed. This duplex can be used as a new type of antisense molecule to pair with an RNA pyrimidine target sequence within an mRNA molecule. This duplex can also be used as a new type of antigene molecule to pair with a single-stranded DNA pytimidine target sequence within the genome.

La présente invention concerne un procédé permettant d'obtenir des duplexes de pyrimidine-purine appariés de Hoogsteen, soit en chauffant un triplex pour dissocier un brin de pyrimidine apparié de Watson-Crick, soit en liant deux brins parallèles au niveau de leur extrémité 5'. Ce duplex peut être utilisé comme un nouveau type de molécule antisens pour former une paire avec une séquence cible pyrimidique d'ARN dans une molécule d'ARNm. De plus, ce duplex peut être utilisé comme un nouveau type de molécule antigène pour former une paire avec une séquence cible pyrimidique d'ADN à simple brin dans le génome.
