Amongst risk factors known to be involved in the pathophysiology of Breast Cancer, estrogenic steroids have been reported to play a major role in the early onset and disease progression. In addition, recent clinical reports claim an increased incidence of AIDS in young women in comparison to any other group of people regardless of race or mode of exposure to HIV virus. We herein report that estrogen can stimulate both NDP-Kinase/Nm23 (EC 2.7.4.6) and Leucine Aminopeptidase (LAPase; EC 3.4.11.1 ) at physiological concentrations leading to an increased activity of both cellular markers. Our results suggest that while estrogen could simultaneously activate NDP-Kinase/Nm23 and LAPase, perhaps leading to higher serum levels of these peptides detected in patients with Breast Cancer, in-vitro studies suggest that estrogen can also enhance HIV infection. We propose that the increased susceptibility to HIV infectivity in cells exposed to estrogen could be related to the effect of estrogen on cellular LAPase activity. The effect of estrogen on LAPase activity resulting in an increased infectivity is in accordance to our previously reported role of this enzyme in HIV viral entry. Finally, the concept of estrogen critical interdependent pathways and their role on Breast Cancer and HIV is discussed.
