A water-insoluble gel suitable as a matrix for electrophoresis, comprising a linear polysaccharide cross-linked with a cross-linker which reacts with hydroxyl groups of the polysaccharide to form other linkages. The cross-linker reaction and gel formation occur simultaneously. The gels have improved optical clarity and give better resolution of small and medium size macromolecules, such as proteins and DNA fragments. The improved resolution occurs at very low total polymer concentrations, in accordance with a new model of gel electrophoresis proposed herein.
