3247-184 ABSTRACT OF THE DISCLOSURE A cross-over linker in single or double-stranded form comprising (a) a homology searching sequence homologous to a targeted base pair sequence of a cloning vehicle; and, (b) a ligatable end complementary to a ligatable end in or insertible into said cloning vehicle, may be used to induce intramolecular cross-over of genetic material and introduce, substitute or delete genetic material In a cloning vehicle. A particular embodiment of the cross-over linker (a "double-cross" linker) has first and second homology searching sequences and can be employed to remove a restriction enzyme site from a cloning vehicle thereby locking up or stabilizing the genetic architective of the cloning vehicle.
