A method of preparing biological materials for cryopreservation is presented. The method lessens the amount of heat released by a cryoprotectant during a latent heat phase by freezing the protectant, thawing the protectant, and treating biologically active materials with the thawed protectant. First, the protectant is frozen to induce an irreversible phase change, along with an irreversible release of energy. After this phase change has occurred, the protectant is thawed and used to treat viable cells or other biologically active material about to undergo freezing. The thawed protectant within the biologically active cells has a reduced endothermic reaction upon subsequent freezing. In one embodiment, freezing the biological material includes immersing the biological material in cooling fluid and circulating the cooling fluid past the biological material at a substantially constant, pre-determined velocity and temperature such that the biological material is vitrified, thus minimizing the formation of stress fractures in cell membranes. The protectant may be propylene glycol, glycerol, DMSO, or other suitable protectants.
Procédé de préparation de matières biologiques en vue d'une cryoconservation. Ce procédé permet de réduire la quantité de chaleur dégagée par un cryoprotecteur pendant une phase de chaleur latente, par la mise en oeuvre des étapes consistant à : congeler l'agent protecteur, le dégeler et traiter les matières biologiques actives à l'aide de l'agent protecteur dégelé. Premièrement, l'agent protecteur est congelé afin d'induire un changement de phase irréversible, ainsi qu'une libération irréversible d'énergie. Une fois que ce changement de phase a eu lieu, l'agent protecteur est dégelé et utilisé pour traiter des cellules viables ou une autre matière biologiquement active devant être congelée. L'agent protecteur dégelé induit une réaction endothermique réduite dans les cellules biologiquement actives, lors d'une congélation ultérieure. Dans un mode de réalisation, la matière biologique comprend les étapes consistant à immerger la matière biologique dans un fluide de refroidissement, à faire circuler ledit fluide sur la matière biologique à une vitesse et à une température prédéterminées sensiblement constantes de manière à vitrifier la matière biologique, ce qui permet de réduire la formation de fractures de contrainte dans les membranes cellulaires. L'agent protecteur peut être du propylène glycol, du glycérol, du DMSO ou d'autres agents protecteurs appropriés.
