The present invention relates to crystals of phosphorylated Aurora-A kinase fragment alone and in complex with a ligand, amino acid residues 1-43 of human TPX2. This invention also relates to methods for designing and selecting ligands, in particular allosteric inhibitors of Aurora-A, that bind to the Aurora-A kinase and their use. La présente invention concerne des cristaux d'un fragment de kinase d'Aurora A phosphorylé seul et dans un complexe avec un ligand, des résidus d'acides aminés 1-43 de TPX2 humain. Cette invention a aussi trait à des méthodes de conception et de sélection de ligands, notamment, d'inhibiteurs allostériques d'Aurora A qui se lient à la kinase d'Aurora A, ainsi qu'à leur utilisation. Par ailleurs, ladite invention a pour objet certains dérivés d'indène et d'indole, des cristaux de kinase d'Aurora A phosphorylée seule et dans un complexe avec un ligand, des résidus d'acides aminés 1-43 de TPX2 humain. L'invention concerne aussi des méthodes de conception et de sélection de ligands se liant à la kinase d'Aurora A, leur utilisation, et un certain dérivé d'indène et d'indole.
