-24- TITLE IMPROVED CULTURE OF HUMAN TUMOR-INFILTRATING LYMPHOCYTES BY COMBINING GAS PERMEABLE BAGS AND A PRIMARY CULTURE CONTAINER ABSTRACT OF THE DISCLOSURE There is disclosed a process for the enhanced expansion and maintenance of human tumor-infiltrating lymphocytes (TIL). The process comprises placing a human tumor sample in a primary culture container and maintaining the sample in the primary culture container for a period of time before transferring a substantial amount of the primary culture to a bulk culture container. The primary culture cells and/or conditioned medium retained in the primary culture container are periodically used to supplement the bulk culture. Both primary and bulk cultures are maintained and expanded by periodic addition of a suitable culture medium as needed.
