The present invention relates to S-phase kinase associated proteins, p19 and p45, referred to herein as "Skp". As described herein, these proteins are components of the tumor cell-specific cyclin A/CDK2 complex and function to function to facilitate DNA replication. Interference with p45 function in vivo prevented entry into S-phase in both normal and transformed cells. Binding data indicated that p45 and p19 associate with each other in a binary complex. Moreover, p45 is required for p19 binding to cyclin A/CDK2.
