The application relates to the purification and characterisation of a family of P-type inositolphosphoglycans (IPGs) from human liver and placenta. These substances are shown to have P-type biological activity, e.g. activating pyruvate dehydrogenase (PDH) phosphatase. The characterisation of the compounds demonstrates that they contain metal ions, in particular Mn2+ and/or Zn2+, and optionally phosphate. The compounds and their antagonists have uses as pharmaceuticals, e.g. for the treatment of diabetes, and in screening for synthetic analogues.
L'invention concerne la purification et la caractérisation d'une famille de phosphoglycans d'inositol (IPG) de type P, obtenus à partir du foie et du placenta humains. On montre que ces substances ont une activité biologique de type P qui consiste, par exemple, à activer la phosphatase de pyruvate-déshydrogénase (PDH). La caractérisation de ces composés démontre qu'ils contiennent des ions métalliques, en particulier Mn?2+¿ et/ou Zn?2+¿ et, en option, du phosphate. Les composés et leurs antagonistes peuvent s'utiliser comme des médicaments, par exemple dans le traitement du diabète et dans la sélection d'analogues synthétiques.
