The present invention is directed to synthetic cytoskeletal active compounds that are related to natural Latrunculin A or Latrunculin B. The present invention is also directed to pharmaceutical compositions comprising such compounds and a pharmaceutically acceptable carrier. The invention is additionally directed to a method of preventing or treating diseases or conditions associated with actin polymerization. In one embodiment of the invention, the method treats increased intraocular pressure, such as primary open-angle glaucoma. The method comprises administering to a subject a therapeutically effective amount of a cytoskeletal active compound of Formula I or II, wherein said amount is effective to influence the cytoskeleton, for example by inhibiting actin polymerization.
L'invention concerne des composés actifs cytosquelettiques synthétiques associés à la latrunculine A ou à la latrunculine B naturelles. L'invention concerne également des compositions pharmaceutiques comprenant lesdits composés et un support acceptable sur le plan pharmaceutique. L'invention concerne en outre un procédé de prévention ou de traitement de maladies ou d'états associés à la polymérisation de l'actine. Dans un mode de réalisation de l'invention, le procédé traite la pression intra-oculaire accrue, notamment le glaucome primitif à angle ouvert. Ce procédé consiste à administrer à un sujet une dose efficace sur le plan thérapeutique d'un composé actif cytosquelettique de formule I ou II, ladite dose étant efficace pour avoir une influence sur le cytosquelette, par exemple par inhibition de la polymérisation de l'actine.
