A cytotoxic agent comprising one or more modified doxorubicins/daunorubicin linked to a cell binding agent. L'invention concerne un agent cytotoxique comprenant une ou plusieurs doxorubicines/daunorubicines modifiées et liées à un agent de liaison cellulaire. Elle concerne également une composition thérapeutique destinée à tuer des populations de cellules choisies, et comprenant: (A) une dose cytotoxique d'au moins une doxorubicine/daunorubicine modifiée et liée de manière covalente à un agent de liaison cellulaire par l'intermédiaire d'un groupe de liaison, et (B) un excipient acceptable sur le plan pharmacologique. L'invention concerne encore un procédé destiné à tuer des populations de cellules choisies et consistant à mettre en contact des cellules cibles ou un tissu contenant des cellules cibles avec une dose efficace d'un agent cytotoxique comprenant au moins une doxorubicine/daunorubicine modifiée et liée à un agent de liaison cellulaire. L'invention concerne enfin des nouvelles doxorubicines/daunorubicines modifiées et contenant du soufre.
