The present invention is directed to the isolation of nucleic acid molecules which encode the enzyme D-gluconolac- tone oxidase (D-GLO) useful in the production of erythorbic acid by the conversion of D-gluconolactone. Various modifications of such nucleic acid molecules are contemplated including the encoded proteins which retain the enzymatic activity of naturally occur- ing D-GLO. Recombinant methods of producing D-gluconolactone oxidase utilizing the nucleic acids of this invention in various host cells transformed by appropriate expression vectors are preferred. Methods of utilizing the D-GLO of the present invention in processes for the conversion of glucose and specifically the conversion of D- gluconolactone to erythorbic acid are also contemplated.
La présente invention concerne l'isolement de molécules d'acide nucléique codant l'enzyme D-gluconolactone oxydase (D-GLO) utile dans la production d'acide érythorbique par la conversion de D-gluconolactone. Diverses modifications de ces molécules d'acides nucléiques sont envisagées, notamment les protéines codées retenant l'activité enzymatique de D-GLO se produisant naturellement. Des méthodes de production par recombinaison de D-gluconolactone oxydase utilisant les acides nucléiques de cette invention dans diverses cellules hôtes transformées par des vecteurs d'expression appropriés sont préférées. Des méthodes d'utilisation de la D-GLO de la présente invention dans des processus de conversion de glucose et spécifiquement la conversion de D-gluconolactone en acide érythorbique sont également prévues.
