The present invention provides a vaccine that can be administered to a mammal to cause immunoprotection in the mammal against a pathogenic organism that has an immunodominant epitope. The vaccine includes a modified form of the antigen in which the immunodominant epitope is located. In this modified form, the immunodominant epitope is immunodampened by any of a number of techniques. Examples of immunodampening techniques include addition of N- linked glycosylation sites, change of the net charge of the epitope, and substitution with a tolerated sequence. The vaccine also includes a pharmacologically acceptable carrier.
L'invention concerne un vaccin qu'on peut administrer à un mammifère, de façon à provoquer l'immunoprotection chez ledit mammifère contre un organisme pathogène possédant un déterminant antigénique immunodominant. Le vaccin comprend une forme modifiée de l'antigène dans lequel est situé le déterminant antigénique immunodominant. Dans cette forme modifiée, le déterminant antigénique est immunoatténué au moyen d'une parmi différentes techniques. Des exemples de techniques d'immunoatténuation comprennent l'apport de sites de glycosylation à liaison N, la modification de la charge nette sur le déterminant antigénique et la substitution par une séquence tolérée. Le vaccin comprend également un véhicule acceptable pharmacologiquement.
