Nucleic acid probes that are susceptible to chemical or enzymatic degradation are described herein. In addition, assays and methods using such probes in the detection of target nucleic acid sequences are disclosed. The target-specific hybridization region or target-complementary region of the degradable probes can be separated via a degradation process from the detectable region. The remaining portion of the degradable probes can be easily detected. The use of the degradable probes described herein improves the signal-to-noise ratio by reducing background specific or non-specific signal generation in assays and methods of nucleic acid detection.
Cette invention concerne des sondes d'acides nucléiques qui sont sensibles à une dégradation chimique ou enzymatique. Sont également décrits des essais et des méthodes qui font intervenir de telles sondes pour la détection de séquences d'acides nucléiques cibles. Dans les sondes dégradables, la région d'hybridation propre à la cible ou complémentaire de la cible peut être séparée de la région détectable par un processus de dégradation. La partie restante des sondes dégradables peut être facilement détectée. L'emploi des sondes dégradables décrites dans cette invention permet d'améliorer le rapport signal-bruit en réduisant la production de signaux de fond spécifiques ou non spécifiques dans le cadre des essais et procédés de détection d'acides nucléiques.
