Methods for the selection of novel signaling allosteric DNA enzymes are provided. In particular, fluorescent signaling allosteric DNA enzymes are described. The selection system is based on the cleavage of an ribonucleotide flanked by a fluorophore modified nucleotide and a quencher modified oligonucleotide. Both cis-acting and trans-acting allosteric DNA enzymes are identified, as well as aptamer/DNA enzyme conjugates.
L'invention concerne des procédés de sélection de nouvelles enzymes allostériques d'ADN de signalisation. L'invention concerne plus particulièrement des enzymes allostériques d'ADN de signalisation fluorescentes. Le système de sélection s'inspire du clivage ribonucléotidique, ribonucléotide venant entre un nucléotide modifié de fluorophore et un oligonucléotide extincteur modifié. On identifie ensuite les deux enzymes allostériques d'ADN sis-actives et trans-actives, ainsi que les conjugués d'enzymes d'aptamère/ADN.
