The growth hormone supergene family comprises greater than 20 structurally related cytokines and growth factors. A general method is provided for creating site-specific, biologically active conjugates of these proteins. The method involves adding cysteine residues to non-essential regions of the proteins or substituting cysteine residues for non-essential amino acids in the proteins using site-directed mutagenesis and then covalently coupling a cysteine-reactive polymer or other type of cysteine-reactive moiety to the proteins via the added cysteine residue. Disclosed herein are preferred sites for adding cysteine residues or introducing cysteine substitutions into the proteins, and the proteins and protein derivatives produced thereby.

Cette invention se rapporte à un procédé général de création de conjugués biologiquement actifs de ces protéines, spécifiques de sites. Ledit procédé consiste à ajouter des restes de cystéine à des régions non essentielles de ces protéines ou à remplacer des restes de cystéine par des acides aminés essentiels dans lesdites protéines au moyen d'une mutagenèse orientée vers un site particulier, puis à coupler par covalence auxdites protéines un polymère réagissant à la cystéine ou un autre type de fraction sensible à la cystéine, par l'intermédiaire du reste ajouté de cystéine. L'invention se rapporte à des sites préférés pour l'ajout des restes de cystéine ou pour la mise en oeuvre des substitutions de cystéine dans les protéines, ainsi qu'aux protéines et aux dérivés protéiques ainsi produits.
