Description

Modified amino acids and methods for their preparation and use as oral delivery systems for pharmaceutical agents are described. The modified amino acids are preparable by reacting single amino acids or mixtures of two or more kinds of amino acids with an amino modifying agent such as benzene sulfonyl chloride, benzoyl chloride, and hippuryl chloride. The modified amino acids may form encapsulating microspheres in the presence of the active agent under sphere-forming conditions. Alternatively, the modified amino acids may be used as a carrier by simply mixing the amino acids with the active agent. The preferred acylated amino acid carrier is salicyloyl- phenylalanine. The modified amino acids are particularly useful in delivering biologically active agents, e.g., desferrioxamine, insulin or cromolyn sodium, or other agents which are sensitive to the denaturing conditions of the gastrointestinal tract.

La présente invention concerne des acides aminés modifiés et leurs procédés d'élaboration et d'utilisation comme systèmes d'administration orale de principes actifs pharmaceutiques. On prépare ces acides aminés modifiés en faisant réagir des acides aminés pris isolément ou en mélanges de deux ou plusieurs acides aminés avec un agent amino-modificateur tel que le chlorure de benzène-sulfonyle, le chlorure de benzoyle et le chlorure d'hippuryle. Les acides aminés modifiés peuvent constituer des microgranules encapsulantes en présence en présence du principe actif dans des conditions de formation des microgranules. Dans une variante, les acides aminés modifiés peuvent servir d'excipient en étant simplement mélangés avec le principe actif. L'acide aminé acylé préféré est le salicyloyle-phénylalanine. Ces acides aminés modifiés sont particulièrement utiles pour l'administration de principes biologiquement actifs tels que la déféroxamine, l'insuline ou le cromolyne sodique, ou d'autres principes sensibles aux conditions dénaturantes du tube digestif.
