The present invention provides a method of reducing the activity of prions using vibriolysin or variants thereof. Vibriolysin-containing solutions are used to sanitize prion-contaminated facilities and instruments and decontaminate food products and bilogical tissues. The present invention provides a method of treating prion-related disease in animals and humans, comprising the administration of a formulation of vibriolysin or a variant thereof together with a pharmaceutically acceptable carrier. Such novel formulations are engineered to track the natual path of the prion from cells where the prions accululate in the preclinical stage into neuronal cells and the brain at the advaced stage of the disease. The present invention provides methods and formulations that encompasses natural and recombinant vibriolysins and variants thereof with enhanced ability to access prion target cells, and with enzyme activity capable of being regulated by specific conditions, such as pH range or enzymatic cleavage.
L'invention concerne un procédé pour diminuer l'activité de prions au moyen de vibriolysine ou de variantes de celle-ci. Des solutions contenant de la vibriolysine sont utilisées pour assainir des milieux et des instruments contaminés par des prions et pour décontaminer des produits alimentaires et des tissus biologiques. La présente invention porte également sur une méthode de traitement de maladies associées aux prions chez les animaux et les humains, cette méthode consistant à administrer une préparation de vibriolysine ou d'une variante de celle-ci avec un excipient pharmaceutiquement acceptable. Ces nouvelles préparations sont mises au point pour suivre la voie naturelle du prion à partir de cellules où les prions s'accumulent à un stade préclinique, jusqu'aux cellules neuronales et au cerveau à un stade avancé de la maladie. Cette invention concerne aussi des procédés et des préparations qui renferment des vibriolysines naturelles et recombinantes, ainsi que des variantes de celles-ci, qui présentent une meilleure aptitude pour accéder aux cellules cibles de prion et une activité enzymatique pouvant être régulée par des conditions spécifiques, telles que le clivage enzymatique ou dans une gamme de pH.
