ABSTRACT A method for detecting micro quantites of nitrogen or sulfur mustards by their conversion to anticholinester- ases is described. The method comprises: reacting an organophosphorus thioacid salt solution with an unknown solution for sufficient time to form compounds having anticholinesterase activity in the reaction solution; contacting the reaction solution with a cholinesterase affixed to a suitable inert support for a time sufficient to inhibit the cholinesterase; adding indoxyl acetate, a substrate for the cholinesterase which, upon hydrolysis in the presence of mild oxidizing agents, produces the insoluble colored material, indigo blue; allowing suf- ficient time for hydrolysis of substrate; and observing the inert support for development of a blue color from preciptation of insoluble indigo blue upon it, or, conversely, for lack of color development. The absence of color indicates the formation of compounds with anti- cholinesterase activity and is a positive test for the presence of the suspect materials. Development of a blue color indicates there was no formation of compounds with anticholinesterase activity and that the suspect materials are not present in the unknown solution or are only present in amounts below the cited limits of sensi- tivity.
