The present invention provides a method of detecting antigens, which comprises immobilizing an antigen to a solid support and contacting the solid support with a means for hybridizing a labeled dendrimer to the antibody, through an oligonucleotide complexed thereto. A directly oligonucleotide labeled primary antibody or an oligonucleotide labeled secondary antibody may be employed, and a conventionally labeled dendrimer can subsequently be hybridized to the oligonucleotide through one or more of the outer arms of the dendrimer. The present invention offers the advantage over conventional methods of antigen detection by providing multiple label molecules per antigen, thereby enhancing the observed signal associated with the label.
La présente invention concerne un procédé permettant de détecter des antigènes et consistant d'abord à immobiliser un antigène sur un support solide, puis à mettre en contact le support avec un dispositif permettant d'obtenir l'anticorps par hybridation d'un dendrimère, la mise en contact se faisant par l'intermédiaire d'un oligonucléotide formant un complexe avec le dispositif. Il est possible d'employer, soit un anticorps primaire directement marqué par l'oligonucléotide, soit un anticorps secondaire marqué par l'oligonucléotide. En outre, un dendrimère marqué de façon conventionnelle peut s'hybrider pour donner l'oligonucléotide via une ou plusieurs des ramifications extérieures du dendrimère. Par rapport aux procédés conventionnels de détection d'antigènes, l'avantage de la présente invention est de donner plusieurs molécules de marquage par antigène, ce qui renforce le signal observé s'associant au marqueur.
