Methods are disclosed for detecting one or more mutations in an isolated test nucleic acid for forming a heteroduplex with a homologous control DNA and contacting the heteroduplex with a resolvase capable of recognizing at least one single base pair mismatch within the heteroduplex. In preferred embodiments of the invention, the resolvase is bacteriophage T4 endonuclease VII.
