The present invention provides compositions, apparatuses and methods for detecting one or more nucleic acid targets present in a sample. Methods of the invention include utilizing two or more oligonucleotide probes that reversibly bind a target nucleic acid in close proximity to each other and possess complementary reactive ligation moieties. When such probes have bound to the target in t proper orientation, they are able to undergo a spontaneous chemical ligation reaction that yields a ligated oligonucleotide product. In accordance with the invention, the presence of the target(s) of interest can be determined by measuring the presence or amount of ligated oligonucleotide product.

La présente invention concerne des compositions, des appareils et des méthodes de détection d'une ou plusieurs cibles d'acides nucléiques présentes dans un échantillon. Les méthodes de l'invention consistent à utiliser deux ou plusieurs sondes oligonucléotidiques se liant de manière réversible à un acide nucléique cible en proximité étroite les unes par rapport aux autres, et possédant des fractions de ligature réactives complémentaires. Une fois liées à la cible selon une orientation t appropriée, ces sondes peuvent subir une réaction de ligature chimique spontanée permettant d'obtenir un produit oligonucléotidique ligaturé. Selon l'invention, la présence d'une ou plusieurs cibles d'intérêt peut être déterminée par mesure de la présence ou de la quantité de produit oligonucléotidique ligaturé.
