The present invention provides methods for producing a nucleic acid fingerprint as well as methods for detecting sequence polymorphisms between one or more genomes. The methods involve: (a) fragmenting a nucleic acid into nucleic acid fragments with ends that are compatible to ligation with at least one adapter, (b) performing a ligation reaction between the compatible ends of the nucleic acid fragments and at least one adapter, (c) amplifying the nucleic acid fragments by using at least one amplification primer, and (d) generating a nucleic acid fingerprint from the amplified fragments. A method according to the present invention permits high-resolution fingerprinting while maintaining stringency in a PCR reaction. In another aspect, the invention also provides a kit for preparing nucleic acid fingerprints according to methods of the invention.
la présente invention concerne des méthodes de production d'empreintes d'acide nucléique ainsi que des méthodes de détection de polymorphismes de séquences au sein d'un génome ou entre plusieurs génomes. Ces méthodes consistent à: (a) subdiviser un acide nucléique en fragments dont les extrémités peuvent former une ligature avec au moins un adapteur; (b) réaliser une réaction de ligature entre les extrémités compatibles de fragments d'acide nucléique et au moins un adapteur; (c) amplifier les fragments d'acide nucléique à l'aide d'au moins une amorce d'amplification; et (d) produire une empreinte d'acide nucléique à partir des fragments amplifiés. Un procédé selon la présente invention permet de réaliser des empreintes à haute résolution sans compromettre la stringence de la réaction en chaîne de la polymérase. Selon un autre aspect, l'invention concerne également un kit pour l'obtention d'empreintes d'acide nucléique.
