The present invention provides methods for the isolation, identification, and purification of adrenomedullin (AM)-binding proteins. Also, provided are methods for utilizing the purified AM-binding proteins, or functional portions thereof, to diagnose, treat, and monitor AM-related diseases, for example, diseases or disorders associated with abnormally elevated AM levels. In addition, the present invention provides a newly identified complex between AM and a specific AM-binding protein 1 (AMBP-1); which has been isolated and identified herein as factor H (fH). The invention also provides AM/AMBP complexes, particularly AM/fH complexes, and antibodies specifically reactive with these complexes. Further provided are methods for identifying and purifying complexes of AM and an AM binding protein using anti-AM/fH antibodies, and methods for treating conditions such as cancer or diabetes utilizing compositions comprising these antibodies. The present invention additionally provides methods for identifying antagonists agents that inhibit the function of AM, factor H, or the AM/factor H complex. The invention also provides methods for treating conditions such as cancer or diabetes using these antagonist agents.

La présente invention porte sur des procédés d'isolation, d'identification et de purification des protéines se liant à l'adrénomédulline (AM). L'invention porte également sur des procédés d'utilisation des protéines de liaison à AM purifiées ou de leurs parties fonctionnelles en vue de diagnostiquer, traiter et surveiller des pathologies apparentées à AM, par exemple, des pathologies ou troubles associés à des taux anormalement élevés en AM. Cette invention porte, de plus, sur un complexe récemment identifié entre AM et une protéine 1 spécifique se liant à AM (AMBP-1); cette protéine a été isolée et identifiée comme étant le facteur H (fH). L'invention porte en outre sur des complexes AM/AMBP, notamment des complexes AM/fH et sur des anticorps réagissant spécifiquement à ces complexes. L'invention porte encore sur des procédés d'identification et de purification des complexes d'AM et d'une protéine de liaison à AM utilisant des anticorps anti-AM/fH et sur des procédés de traitements d'états tels que le cancer ou les diabètes utilisant des compositions comprenant ces anticorps.
