A method and kit for measurement of a steroid by means of a competitive immunoassay, preferably a competitive enzyme immunoassay. The method and kit involve the use of a steroid analogue conjugated to a label. The steroids that are amenable to detection by the method and kit of the present invention include estradiol and progesterone. The method comprises the steps of: a) incubating a mixture of a test sample suspected of containing a given steroid, a solid phase coupled to an antibody specific for that steroid, and a conjugate of an analogue of that steroid to form steroid/antibody complexes and conjugate/antibody complexes on said solid phase; b) separating said solid phase from said mixture; c) measuring the amount of label present in said mixture or in said solid phase; and d) determining the amount of steroid in said sample from the amount of label. The kit comprises a solid phase coupled to an antibody specific for a steroid and a conjugate of an analogue of that steroid.
Cette invention concerne un procédé, et le kit correspondant, permettant la mesure d'un stéroïde au moyen d'un dosage immunologique par compétition, de préférence un dosage immunologique par compétition faisant intervenir des enzymes. Ce procédé, ainsi que le kit correspondant, consiste en l'utilisation d'un analogue de stéroïde conjugué à un marqueur. Parmi les stéroïdes qui peuvent faire l'objet de la détection par ce procédé et au moyen du kit correspondant décrit dans l'invention, on compte l'oestradiol et la progestérone. Ledit procédé consiste (a) à faire incuber un mélange d'un échantillon test suspecté de contenir un stéroïde donné, d'une phase solide couplée à un anticorps spécifique de ce stéroïde, et d'un conjugué d'un analogue de ce stéroïde pour former des complexes stéroïde/anticorps et conjugué/anticorps sur ladite phase solide, (b) à séparer ladite phase solide dudit mélange, (c) à mesurer la quantité de marqueur présent dans ledit mélange ou dans ladite phase solide, et (d) à déduire la quantité de stéroïdes dans ledit échantillon à partir de la quantité de marqueur présent. Le kit de l'invention comporte une phase solide couplée à un anticorps spécifique d'un stéroïde, et un conjugué d'un analogue de ce stéroïde.
