A method for determining whether a microorganism produces an AmpC .beta.~lactamase is disclosed in which a culture of a microorganism suspected of producing a .beta.-~lactamase that inactivates a .beta.-lactam-containing antibiotic is admixed with an effective amount of each of i) a .beta.-lactam- containing antibiotic, ii) a .beta.-lactamase inhibitor to which AmpC .beta.- ~lactamase is resistant, and iii) a permeabilizing agent for the microorganism present in a non~-growth-inhibiting microorganism-permeabilizing amount to form an assay culture. That assay culture in maintained under appropriate culture conditions and for a time period sufficient to determine the interaction of the microorganism with the AmpC .beta.-lactamase resistant inhibitor and antibacterial compound, and thereby determine the presence of an AmpC .beta.-lactamase, wherein a positive test indicates the presence of an AmpC .beta.-lactamase.
L'invention concerne un procédé permettant de déterminer si un micro-organisme produit une AmpC ß-lactamase. Selon ce procédé, une culture de micro-organismes suspectés de produire une ß-lactamase qui inactive un antibiotique contenant une ß-lactame est mélangé à une quantité efficace de: i) un antibiotique contenant une ß-lactame, (ii) un inhibiteur de ß-lactamase auquel l'AmpC ß-lactamase est résistante, et (iii) un agent perméabilisant pour le micro-organisme présent dans une quantité perméabilisante de micro-organismes inhibant une non-croissance, de sorte à former une culture de dosage. Cette culture de dosage est maintenue dans des conditions de culture appropriées, pendant une durée suffisante pour déterminer l'interaction du micro-organisme avec l'inhibiteur résistant à l'AmpC ß-lactamase et un composé antibactérien, ce qui permet de déterminer la présence d'une AmpC ß-lactamase, un test positif indiquant la présence de cette enzyme.
