A method and device to assist in the determination of protein domain boundaries is described. In particular the device is designed to provide a high throughput of proteolytic digestion of proteins to identify domains and their boundaries, for use in protein structure determination, in a manner that is amenable to automation. Proteases are immobilized in a convenient format such as a microtitre plate and preferably arranged in a matrix thereby allowing for simultaneous degradation of a protein by a number of proteases at a number of concentrations.
L'invention concerne un procédé et un appareil permettant de déterminer les limites d'un domaine de protéine. Cet appareil est conçu, en particulier, pour fournir aux protéines une capacité de digestion protéolytique élevée permettant d'identifier des domaines et leurs limites. Le procédé est utilisé pour déterminer une structure protéinique, de telle manière qu'il peut être automatisé. Les protéases sont immobilisées dans un format approprié tel qu'une plaque de microtitre, et de préférence placées dans une matrice, ce qui permet une dégradation simultanée d'une protéine par un certain nombre de protéases, selon un certain nombre de concentrations.
