A device for conducting integrated sequential separation and enrichment of a mixture of sample proteins, comprising means for separation and means for enrichment, whereby free flow electrophoresis - isoelectric focussing (FFE- IEF), is arranged to take place within an etched system of basins and channels is preferably employed in the separation step, multichannel conduits are used to guide the separated protein fractions to the enrichment stage, and a solid phase micro-extraction procedure, in an optionally dockable format, is preferably employed in the enrichment step. The separated fractions are then preferably dispensed onto a MALDI-plate for subsequent matrix assisted laser desorption ionisation (MALDI) analysis.

L'invention porte sur un dispositif permettant de mener à bien la séparation séquentielle intégrée et l'enrichissement d'un mélange de protéines échantillons et comprenant des moyens de séparation et des moyens d'enrichissement. A cet effet, une focalisation isoélectrique-électrophorèse à écoulement libre a lieu dans un système gravé de bassins et de canaux que l'on utilise de préférence lors de la séparation. Des conduits multicanal sont utilisés pour guider les fractions de protéines séparées vers l'étage d'enrichissement, et un procédé de micro-extraction en phase solide, éventuellement ancrable, est utilisé de préférence à l'étape d'enrichissement. Les fractions séparées sont ensuite distribuées de préférence sur une plaque MALDI en vue d'une analyse par désorption-ionisation laser assistée par matrice (MALDI).
