The invention relates to a device for presenting peptides or proteins which can be used as a polypeptide chip for miniaturized detection of molecules which structurally or functionally complement said polypeptides. Said device consists of a flat support where polypeptides are covalently bound. The link between the polypeptides and the support results from the formation of a semicarbazone link. The semicarbazone link is a specific result of the reaction between polypeptides having an aldehyde or ketone function; and a support functionalized by semicarbazide groupings. The invention also relates to a method for preparing the supports and attaching the polypeptides to these supports in addition to the use of said devices thus obtained as polypeptide chips .
L'invention concerne un dispositif de présentation de peptides ou de protéines, qui est utilisable comme "puce à polypeptides" pour la détection miniaturisée de molécules structurellement ou fonctionnellement complémentaires desdits polypeptides. Ce dispositif consiste en un support plat sur lequel les polypeptides sont liés de manière covalente, cette liaison entre le polypeptides et le support résultant de la formation d'un lien semicarbazone. Le lien semicarbazone résulte en particulier de la réaction entre- des polypeptides portant une fonction aldéhyde ou cétone et son support fonctionnalisé par des groupements semi-carbazides. L'invention concerne également le procédé de préparation des supports et de fixation des polypeptides sur ces supports ainsi que l'utilisation des dispositifs ainsi obtenus comme puces à polypeptides.
