By this invention, acyltransferase proteins are provided capable of catalyzing the production of triglycerides from 1,2-diacylglycerol and an acyl-CoA. The invention comprises a partially purified diacylglycerol acyltransferase (DAGAT), wherein said protein is active in the formation of triacylglycerol from fatty acyl-CoA and diacylglycerol substrates. Of special interest is a Mortierella ramanniana DAGAT having a molecular mass of approximately 40kD. Also considered are amino acid and nucleic acid sequences obtainable from DAGAT proteins and the use of such sequences to provide transgenic host cells with modified triacylglycerol levels.
La présente invention se rapporte à des protéines acyltransférase capables de catalyser la production des triglycérides à partir de 1,2-diacylglycérol et d'un acyl-coA. L'invention comprend un diacylglycérol acyltransférase (DAGAT) partiellement purifié, ladite protéine étant active dans la formation du triacylglycérol à partir d'un acyl-CoA gras et de substrats de diacylglycérol. Un élément particulièrement intéressant est une DAGAT Mortierella ramanniana dont la masse moléculaire est d'environ 40KD. L'invention se rapporte également à des séquences d'acides aminés et d'acides nucléiques pouvant être obtenues à partir des protéines DAGAT, ainsi qu'à l'utilisation de telles séquences pour obtenir des cellules hôtes transgéniques avec des taux de triacylglygérol modifiés.
