Isolated, monoclonal, human, anti-/3-amyloid antibodies are provided which bind to dimeric forms of Ab with higher affinity than to monomelic forms of Ab and when bound to an A.beta. polypeptide comprising A.beta.(21 -37) shield A.beta.(21-37) from proteolytic digestion. The antibodies were shown to inhibit fibril formation and reduce plaque size in vivo and to not bind brain vessel walls. Accordingly, the antibodies are useful in human and veterinary medicine for the treatment and prophylaxis of Alzheimer's disease and other neurodementing diseases. Methods of detecting or measuring the progression of a neurodementing disease also are provided.
L'invention porte sur des anticorps isolés, monoclonaux, humains, anti-/3-amyloïde qui se fixent à des formes dimères d'Ab avec une affinité plus forte qu'aux formes monomères d'Ab, quand ils sont fixés à un polypeptide Aß comprenant la protection Aß (21-37) de la digestion protéolytique. Ces anticorps ont montré qu'ils inhibaient la formation de fibrilles et réduisaient la taille des plaques in vivo, et cela sans se fixer aux parois des vaisseaux du cerveau. Ces anticorps sont donc utiles en médecine humaine et vétérinaire pour le traitement et la prophylaxie de la maladie d'Alzheimer, et de maladies neurodémentielles. L'invention porte également sur des méthodes de détection et mesure de la progression de maladies neurodémentielles.
