The invention provides an improved test cell for detecting the presence of an analyte in a liquid sample. The device has an elongate casing (10) defining a liquid sample inlet, a reservoir volume a test volume and a window (16) through the casing at the test volume. Disposed within the cell is a sample absorbent, a novel biphasic substrate and a reservoir, together capable of transporting an aqueous solution within the casing along a flow path extending from the sample inlet through the test volume and into the reservoir volume. The invention further comprises a method for detecting the presence of an analyte in a liquid sample using the device and a biphasic chromatographic material for carrying out the method. The release medium is e.g. absorbent paper and holds a band of e.g. antibody-metal sol, and a band of e.g. antibody- biotin which bind to first and second epitopes respectively of the analyte. The capture site is e.g. nitrocellulose or nylon, holding an immobilized capture component, e.g. streptavidin. A control site may be present. The release medium and capture site are joined by overlapping to form the chromatographic substrate.
L'invention se rapporte à une cellule d'analyse améliorée qui permet de déceler la présence d'un analysat dans un échantillon de liquide. Ce dispositif comporte un boîtier allongé (10) qui définit un orifice d'admission d'échantillon de liquide, un volume tenant lieu de réservoir, un volume pour l'analyse, et une fenêtre (16) pratiquée dans le boîtier au niveau du deuxième volume d'analyse. La cellule contient un absorbant d'échantillon, un substrat biphasique nouveau et un réservoir, capables ensemble d'introduire une solution aqueuse dans le boîtier par écoulement dans un passage qui s'étend depuis l'orifice d'admission des échantillons, traverse le volume réservé à l'analyse et aboutit dans celui du réservoir. L'invention concerne également un procédé qui permet de déceler la présence d'un analysat dans un échantillon de liquide en utilisant ce dispositif et un matériel chromatographique biphasique de mise en oeuvre du procédé. Le support de libération est, par exemple, un papier absorbant et il contient une bande, par exemple, d'anticorps-métal sol, ainsi qu'une bande, par exemple, d'anticorps-biotine qui se lie aux premier et second épitopes respectivement de l'analysat. Le site de capture est, par exemple, de la nitrocellulose ou du nylon, contenant un constituant de capture immobilisé, par exemple, le streptavidine. On peut prévoir la présence d'un site témoin. Le support de libération et le site de capture sont joints par chevauchement pour former le substrat chromatographique.
