The present invention relates to a method for detecting one or more nascent RNAs in a tissue sample using FISH. In the method, a plurality of probes (8 to 82) may be used to detect a single species of nascent RNA. Further, a plurality of nascent RNA species may be detected simultaneously using between 8 and 82 probes for each nascent RNA. The invention comprises, in addition, methods of preparing a sample for nascent RNA detection by reducing autofluorescence of the tissue sample. These techniques may be synergistically combined to achieve significantly improved results.

L'invention concerne un procédé permettant de détecter au moins un ARN naissant dans un échantillon tissulaire par hybridation in situ en fluorescence (FISH). Dans ce procédé, on peut utiliser plusieurs sondes (8 à 82) pour détecter une espèce simple d'ARN naissant. En outre, on peut détecter simultanément plusieurs espèces d'ARN naissants en utilisant 8 à 82 sondes pour chaque ARN naissant. L'invention concerne en outre des procédés qui permettent de préparer un échantillon pour la détection d'ARN naissant par réduction de l'autofluorescence de l'échantillon tissulaire. On peut combiner ces techniques de manière synergique pour obtenir des résultats sensiblement meilleurs.
