With respect to a range of 1.5 Mbp and more in the chromosome 19p13.3 region containing Peutz-Jeghers gene, a continuous cosmid contig is constructed and a restriction map is prepared. Next, genes mapped within this region are searched by using EST database and the locations of these genes are accurately determined. Based on the evaluation of biological data, etc., several highly likely candidates for Peutz-Jeghers gene are specified from the genes thus found. After successively analyzing variations in these genes in DNAs of patients with Peutz-Jeghers syndrome, it is found that one of these genes, i.e., "LKB1" has been specifically varied in these patients. Thus, it is found out that the diseases caused by the variation in the LKB1 gene can be examined and treated by using the LKB1 gene, primers and probes based on its base sequence, LKB1 protein, an antibody binding to this protein, etc.
Compte tenu d'une gamme de 1,5 Mbp et plus dans la région chromosomique 19p13.3 contenant le gène de Peutz-Jeghers, on construit un contraction de clones contigus continue de cosmide et élabore une carte de restriction. On recherche ensuite les gènes cartographiés dans cette région en faisant appel à une base de données EST et l'on détermine avec précision les emplacements de ces gènes. D'après cette évaluation de données biologiques plusieurs gènes à candidature hautement probable relativement au gène de Peutz-Jeghers sont spécifiés à partir des gènes ainsi découverts. A la suite d'analyses successives des modifications survenant dans ces gènes dans l'ADN de patients atteints du syndrome de Peutz-Jeghers, on a découvert que l'un de ces gènes, en l'occurrence le gène "LKB1", avait subi des modifications de manière spécifique chez lesdits patients. On a, de la sorte, découvert que les affections provoquées par une modification dans le gène LKB1 pouvait être examinées et traitées à l'aide de ce gène LKB1, d'amorces et de sondes basées sur sa séquence de base, de la protéine de LKB1 et d'un anticorps se fixant à cette protéine, etc.
