This invention relates to the protein, Mycobacterium paratuberculosis acylase (mpa) and the gene encoding mpa, which we have identified in the pathogen Mycobacterium paratuberculosis Mptb (also designated Mycobacterium avium subspecies paratuberculosis MAP), and to their use in the diagnosis of Mptb/MAP infections in animals and humans, as well as their use as components of vaccines for the prevention and treatment of diseases caused by Mptb/MAP. The importance of an intact uninterrupted mpa gene as a determinant of pathogenicity in Mptb/MAP is recognised and the invention also provides attenuated strains of normally pathogenic Mptb/MAP and other mycobacteria in which mpa has been inactivated, for use as vaccines.
Cette invention se rapporte à la protéine, Mycobactérium paratuberculosis acylase (mpa) et au gène codant cette protéine mpa, qui ont été identifiés dans l'agent pathogène Mycobactérium paratuberculosis Mptb (également appelé Mycobactérium avium sous-espèce paratuberculosis MAP) et à leur utilisation dans le diagnostic des infections par Mptb/MAP, chez des animaux et chez l'homme, ainsi qu'à leur utilisation comme constituants de vaccins pour la prévention et le traitement des maladies causées par Mptb/MAP. L'importance d'un géne de mpa ininterrompu et intact comme déterminant du pouvoir pathogène de Mptb/MAP est reconnue et cette invention fournit également des souches atténuées de Mptb/MAP normalement pathogène et d'autres mycobactéries dans lesquelles la protéine mpa a été inactivée, en vue de leur utilisation comme vaccins.
