Asymmetric derivatives of furamidines with one of the phenyl rings of furamidine replaced with a benzimidazole have been found by quantitative footprinting analyses to bind GC containing sites on DNA more strongly than to pure AT sequences. These compounds have been shown to bind in the minor groove at specific GC containing sequences of DNA in a highly cooperative manner as a stacked dimer. Compounds of the present invention find use in selectively binding mixed sequence DNA, and may also be used in methods of regulating gene expression, methods of treating opportunistic infections and cancer, as well as in methods of detecting certain sequences of DNA.

On a découvert par des analyses d'empreintes quantitatives que des dérivés asymétriques de furamidines, dans lesquels l'un des cycles phényle du furamidine est remplacé par un benzimidazole, se fixent aux sites contenant des bases GC sur l'ADN, de façon beaucoup plus forte que sur des séquences AT pures. Ces composés ont révélé qu'ils se fixent dans la petite rainure de l'ADN au niveau de séquences contenant des bases GC spécifiques d'une manière hautement coopérative, sous la forme d'un dimère empilé. De tels composés trouvent une application dans la fixation sélective de séquences mixtes d'ADN et ils peuvent être utilisés dans des procédés de régulation de l'expression génique, dans des procédés de traitement des infections opportunistes et du cancer, ainsi que dans des procédés de détection de certaines séquences d'ADN.
