A diaphragm valve including a housing forming an open chamber, and inlet and outlet conduits opening into such chamber. A flexible membrane is mounted in the chamber and operated by a control valve to move the flexible membrane between an open position permitting fluid flow through the valve and a closed position blocking such flow. In some embodiments, the membrane is moved to its open position by exposing the upper surface of the membrane to a negative pressure in the outlet conduit and utilizing atmospheric pressure to close the valve. In other embodiments, a positive pressure is used to close the valve, and atmospheric pressure permits the membrane to move to its open position. In some embodiments, the membrane may be provided with a stiffening member so that it can seat across the opening of the outlet conduit in its closed position. In some embodiments, the inlet and outlet conduits are axially aligned, and in other embodiments they extend in perpendicular relation to one another.
