The nonstructural protein 4 (NSP4) in the SA11 ATCC rotavirus strain has a histidine at amino acid position 47. This substituted form is more cytotoxic than the NSP4 of the Australia rotavirus strain, which has an asparagine at amino acid position 47. The histidine at amino acid position 47 is mutagenized to another amino acid to produce an alternative form of NSP4 which has reduced toxicity, while retaining its antigenicity and immunogenicity. NSP4 having a glutamic acid at amino acid position 48 is more cytotoxic than NSP4 having a lysine at amino acid position 48. The lysine at amino acid position 48 is mutagenized to another amino acid other than glutamic acid to produce an alternative form of NSP4 which has reduced toxicity, while retaining its antigenicity and immunogenicity.

La protéine 4 non structurale (NSP4) de la souche du rotavirus SA11 ATCC possède une histidine à la position 47 de l'acide aminé. Cette forme substituée est plus cytotoxique que la NSP4 de la souche du rotavirus australienne qui possède une aspargine à la position 47 de l'acide aminé. L'histidine à la position 47 de l'acide aminé est mutagénisée à un autre acide aminé de façon à produire une forme alternative de NSP4 à toxicité réduite, mais conserve son antigénicité et son immunogénicité. NSP4 possédant un acide glutamique à la position 48 est plus cytotoxique que NSP4 possédant une lysine à la position 48 de l'acide aminé. La lysine à la position 48 de l'acide aminé est mutagénisée à un autre acide aminé autre que l'acide glutamique pour produire une forme alternative de NSP4 à toxicité réduite, mais conservant son antigénicité et son immunogénicité.
