A differential expression screening method is provided for identifying a genetic element involved in a cellular process, which method comprises comparing: (a) gene expression in a first cell of interest; and (b) gene expression in a second cell of interest, which cell comprises altered levels, relative to physiological levels, of a biological molecule implicated in the cellular process, due to the introduction into the second cell of a heterologous nucleic acid directing expression of a polypeptide; and identifying a genetic element whose expression differs, wherein gene expression in said first and/or second cell of interest is compared under at least two different environmental conditions relevant to the cellular process.
L'invention concerne un procédé d'analyse par expression différentielle qui permet d'identifier un élément génétique impliqué dans un processus cellulaire. Ce procédé consiste à comparer : (a) l'expression génétique dans une première cellule donnée, et (b) l'expression génétique dans une seconde cellule donnée, cette seconde cellule comprenant des taux modifiés par rapport aux taux physiologiques d'une molécule biologique impliquée dans le processus cellulaire, résultant de l'introduction dans cette seconde cellule d'un acide nucléique hétérologue régulant l'expression d'un polypeptide, puis à identifier un élément génétique dont l'expression est modifiée. L'expression génétique dans la première et la seconde cellule est comparée dans au moins deux types différents de conditions ambiantes se rapportant au processus cellulaire.
