The present invention is based upon the identification of genes which are differentially expressed in hypertrophic cardiac tissue as compared to normal cardiac tissue. Accordingly, the present invention provides nucleotide sequences of genes selected from the group consisting of CH-1, CH-2, CH-3, CH- 4, CH-5, CH-6, CH-7, CH-8, and CH-9, and amino acid sequences of their encoded proteins, as well as derivatives (e.g., fragments) and analogs thereof. The invention also provides therapeutic methods and pharmaceutical compositions which are based on the promotion or inhibition of the function of the differentially expressed genes. The invention further provides methods of diagnosis, prognosis and screening for a disposition for diseases or disorders associated with cardiac hypertrophy. Methods for screening for modulators of the protein products of the differentially expressed genes in cardiac hypertrophy tissue are additionally provided.
Cette invention a trait à l'identification de gènes exprimés de façon différentielle dans le tissu cardiaque hypertrophique en comparaison de ce qu'il en va dans le tissu cardiaque normal. Elle porte sur des séquences nucléotidiques de gènes issus du groupe constitué par CH-1, CH-2, CH-3, CH-5, CH-6, CH-7, CH-8 et CH-9 et sur des séquences aminoacides de leurs protéines codées ainsi que sur des dérivés (des fragments, par exemple) et des analogues de ces séquences. Elle porte, de surcroît, sur des méthodes thérapeutiques et des compositions pharmaceutiques afférentes se fondant sur la stimulation ou l'inhibition de la fonction des gènes exprimés de façon différentielle. Elle a trait, en outre, à des méthodes diagnostiques, pronostiques et de criblage visant à déterminer une propension à des maladies ou à des troubles associés à l'hypertrophie cardiaque. Il y est également traité de méthodes de criblage de modulateurs de produits protéiques de gènes exprimés de façon différentielle dans le tissu cardiaque hypertrophique.
