GPC (gel permeation chromatography) and size exclusion chromatography with detection by multi-angle light-scattering photometry (SEC-MALS) can be used to accurately measure molecular size and molar mass, respectively, of saccharide conjugates. The invention provides (a) a process for measuring the molecular size of a conjugated saccharide antigen within a sample, comprising a step of analysing the sample by GPC, and (b) a process for measuring the molar mass of a conjugated saccharide antigen within a sample, comprising a step of analysing the sample by SEC-MALS.
Selon l'invention, on utilise la chromatographie de perméation sur gel ("gel permeation chromatography" ou GPC) et la chromatographie d'exclusion stérique avec détection par photométrie de diffusion de la lumière multi-angles ("size exclusion chromatography with detection by multi-angle light-scattering photometry" ou SEC-MALS) pour mesurer avec précision la taille et la masse molaire, respectivement, de conjugués du saccharide. L'invention se rapporte à (a) un procédé qui permet de mesurer la taille moléculaire d'un antigène de saccharide conjugué dans un échantillon, lequel procédé comprend une étape d'analyse GPC de l'échantillon, et (b) un procédé qui permet de mesurer la masse molaire d'un antigène de saccharide conjugué dans un échantillon, lequel procédé comprend une étape d'analyse SEC-MALS de l'échantillon.
