1,2-Dioxetane compounds bearing a proteolytic enzyme specific amino acid or peptide are provided, which amino acid or peptide can be removed by action of the corresponding protease. When the amino acid or peptide is removed, the 1,2- dioxetane decomposes with chemiluminescence, the generation of light providing a rapid, ultra-sensitive and convenient means for detecting the presence of the protease in the sample being inspected. The amount of light generated, or degree of chemiluminescence, can be correlated with the amount of protease present. Immunoassays, as well as DNA hybridization and DNA probe assays are provided.
L'invention concerne des composés 1,2-dioxétane portant un acide aminé ou un peptide spécifique à une enzyme protéolytique, lequel acide aminé ou peptide peut être enlevé par l'action de la protéases correspondante. Quand l'acide aminé ou le peptide est enlevé, le 1,2-dioxétane se décompose avec chimiluminescence, la production de lumière constituant un moyen rapide, ultrasensible et commode pour détecter la présence de la protéase dans l'échantillon examiné. La quantité de lumière produite, ou le niveau de chimioluminescence, permet de déduire la quantité de protéase présente. On décrit des dosages immunologiques, ainsi que des tests utilisant l'hybridation de l'ADN et des sondes d'ADN.
