The invention is based on the discobery that the presence of a discordant helix in a protein or peptide is predictive of that protein or peptide's ability to form amyloid. The invention includes methods for detecting discordant helices and methods of screening for compounds that stabilize the I- helix of a discordant helix-containing polypeptide. Compounds discoverd using these methods are useful for treating or preventing disorders in which amyloid is produced. Such disorders include Alzheimer's disease and prion-associated disorders.

L'invention concerne la découverte de la présence d'une hélice discordante dans une protéine ou un peptide permettant de prédire la possibilité de la protéine ou du peptide de former une substance amyloïde. L'invention concerne également des procédés de détection d'hélices discordantes et des procédés de dépistage de composants permettant de stabiliser l'hélice-I d'un polypeptide porteur d'une hélice discordante. Les composants découverts grâce à l'utilisation de ces procédés servent à traiter ou prévenir des troubles dans lesquels la substance amyloïde est produite. Ces troubles comprennent la maladie d'Alzheimer et les troubles associés au prion.
