The present invention relates to a novel method for the amplification of DNA, this method being particularly useful for the amplification of the DNA or the whole genome of a single cell, chromosomes or fragments thereof. Described is also the use of the method in DNA analysis for medical, forensic, diagnostic or scientific purposes, like comparative genomic hybridization (CGH)-, fluorescence in situ hybridization (FISH)-, polymerase chain reaction (PCR)-, single strand conformation polymorphism (SSCP)-, DNA sequence-, "loss of heterozygosity" (LOH)-, fingerprint- and/or restriction fragment length polymorphism (RFLP)- analysis.

La présente invention concerne un procédé d'amplification de l'ADN convenant prticulièrement à l'amplification de l'ADN ou du génome total d'une unique cellule, de chromosomes ou de certains de leurs fragments. L'invention concerne également la mise en oeuvre de ce procédé pour l'analyse d'ADN à des fins médicales, judiciaires, diagnostiques ou scientifiques. Il s'agit notamment d'analyse CGH (hybridation génomique comparative), FISH (hybridation in situ par fluorescence), d'analyse PCR (réaction en chaîne par polymérase), d'analyse SSCP (polymorphisme de conformation monobrin), d'analyse de séquence d'ADN, d'analyse LOH (perte d'hétérozygotisité), d'analyse d'empreinte et/ou d'analyse RFLP (polymorphisme de longueur de fragments de restriction).
