An isolated DNA construct comprising: a) a first DNA sequence comprising either an inducible promoter sequence responsive to the presence or absence of an exogenous inducer or a developmental gene promoter capable of initiating gene expression in a selected tissue or at a selected stage of development of an organism; b) a second DNA sequence comprising a DNA sequence coding for a protease enzyme operably linked and under the control of the promoter sequence specified at (a); whereby the presence or absence of the exogenous inducer or the activation of the developmental gene promoter specified at (a) results in expression of said protease enzyme. These constructs are preferably rendered reversible by the presence of further elements. They can be used in plant or mammalian cells for disruption of cell function, controlling senescence and modifying the metabolism of stored proteins.
L'invention concerne une construction d'ADN sous forme isolée, comprenant: a) une première séquence d'ADN comportant soit une séquence promoteur inductible, sensible à la présence ou à l'absence d'un inducteur exogène, soit un gène promoteur du développement, capable de démarrer l'expression génique dans un tissu choisi, ou à une étape choisie du développement d'un organisme; b) une seconde séquence d'ADN comprenant une séquence d'ADN codant pour une protéase liée de manière fonctionnelle et sous la commande de la séquence promoteur décrite en (a); de sorte que la présence ou l'absence de l'inducteur exogène ou l'activation du gène promoteur du développement décrit en (a) a pour conséquence l'expression de ladite protéase. De préférence, ces constructions sont rendues réversibles par la présence d'autres éléments. On peut les utiliser dans des cellules végétales ou mammifères, aux fins d'interruption de la fonction cellulaire, de maîtrise de la sénescence et de modification du métabolisme de protéines stockées.
